I think you need to grasp some basic concepts:
Disk, including sectors, tracks, clusters, partition tables, file allocation tables, etc.; you can learn them together with using NDD and the like.
Directories and files, including the main filename, extension, attributes; you can learn them together with using PCTOOLS and related commands.
Memory, concepts like UMB, XMS, EMS, etc.; basically learn them together with using MI
Once you learn these concepts, you'll know DOS too.

For beginners, starting right away with disk structure and the like is not good, it's too dull. You should first learn practical use, such as common operations, utility software, and so on. Disk structure should be left for the end. After all, you have to consider what most people can absorb.
