I recently bought a USB hard drive enclosure, with a Seagate 8.0G hard drive inside. Under DOS I got it working successfully through the aspiuhci.sys driver. Does this mean that on other computers I can also use aspiuhci.sys to drive this “big USB flash drive”?

ASPIUHCI is quite sensitive to the model of the USB interface chip... so its compatibility is rather low...

Then if I use it together with aspiehci.sys and aspiohci.sys, would that achieve the goal of being universal?
