Not sure whether everyone uses pirated software, and I don't know if you've used a CD called "Clone Era." When you use it to boot the computer, it first lets you choose whether to boot from the CD-ROM or the hard disk.

Boot from Hard Disk
Boot from CD-ROM

There is a 10-second wait time, and by default it boots from the hard disk

I looked into it but couldn't figure it out

But I noticed there was an extra jo.sys inside, yet when I copied it to another boot disk (a Win98 startup disk), it had no effect. What's going on?

That isn't nu2.com's jo.sys, it's the original DOS one. But please look at the multi-boot method in the latest version of "Super MS-DOS 7.10 Boot Disk":

I want to know, for that boot from hard disk you can use jo.sys, and for GRUB's boot from hard disk you can directly use a statement immediately. So if I don't want to use that jo.sys and want to use DOS to boot directly from the hard disk, can it be done by writing a statement?
