In the DOS article, it is said that use srctool to create the MBR image file bootsect.dos. I want to ask, isn't this created image file still modified by Windows 2000? How can it boot a pure DOS?

Yes, such as using the SYS D: or FORMAT D: /S command. But generally, it won't boot unless you use a multi-boot management program.
