I made a boot disk under DOS 6.22, but it can't boot from drive A (the system says: NON-SYSTEM DISK)
Remove this disk, press Enter, enter DOS 6.22, type a:, press Enter, and then I can enter drive A. Using the dir command
I can also see the boot files inside. If I switch to another boot disk I made, it boots fine. I tried both PCTOOLS and HD-COPY, but it still doesn't work.

What causes this disk to be readable only, but unusable as a boot disk? What method can solve it?

How did you make it? Did you use the FORMAT or SYS command?
There is a DISKTOOL in NU that can replace the DOS system version at will. You can use it to turn that floppy into a system disk.

I first used NU's DISKTOOL, then used NDD to test it, and it was fine (maybe track zero was bad).

In use I found NU is still a bit better than PCTOOLS; you can tell from this point alone, though I'm not sure if that's the case

No wonder NU swallowed up PCTOOL.

DISKTOOLS can be used specifically to solve some problems with disks that can't boot.
But the strongest solution I've tried was installing WIN95/98. I once ran into two hard disks that absolutely would not boot DOS no matter what; after installing WIN98 they worked normally.
